182 ΔNp63-mediated repression of the lncRNA NEAT1 modulates epidermal differentiation

چکیده
The transcription factor ΔNp63, a master regulator of epithelial biology, is involved in regulating stem cell function and maintaining the integrity stratified tissues such as epidermis. To this end, ΔNp63 acts transcriptional repressor or activator towards distinct subset protein-coding genes microRNAs. However, how whether regulates long non-coding RNA (lncRNAs) expression not known. Here, we identified lncRNAs NEAT1 bona fide target. We found that proliferating human keratinocytes represses by recruiting histone deacetylates HDAC1/HDAC2 to p63 DNA binding site located proximal promoter genomic locus. Upon induction differentiation, down-regulation associated marked increase levels, resulting an increased assembly paraspeckles foci both vitro skin tissues. RNA-seq analysis with global profile (ChIRP-seq) revealed associates key factors sustaining their during epidermal differentiation. These molecular events might explain inability NEAT1-depleted undergo correct differentiation program. Remarkably, dysregulated those pathologies characterized aberrant proliferation/differentiation switch ichthyosis psoriasis, suggesting its potential involvement pathogenesis disorders.

ΔNp63 intronic miR-944 is implicated in the ΔNp63-mediated induction of epidermal differentiation
ΔNp63 is required for both the proliferation and differentiation of keratinocytes, but its role in the differentiation of these cells is poorly understood. The corresponding gene, TP63, harbors the MIR944 sequence within its intron. However, the mechanism of biogenesis and the function of miR-944 are unknown. We found that miR-944 is highly expressed in keratinocytes, in a manner that is concor...
متن کاملYAP Expression and Activity Are Suppressed by S100A7 via p65/NFκB-mediated Repression of ΔNp63.
In several squamous cell carcinoma (SCC) cells, it has been previously observed that induction of the S100 calcium-binding protein A7 (S100A7) is repressed by YAP via the Hippo pathway. This report now demonstrates that S100A7 also represses YAP expression and activity by ΔNp63 in cancer cells. Stable overexpression of S100A7 activates the NFκB pathway and inhibits the expression of ΔNp63. Caff...
متن کاملAberrant epidermal differentiation and disrupted ΔNp63/Notch regulatory axis in Ets1 transgenic mice
The transcription factor Ets1 is expressed at low levels in epidermal keratinocytes under physiological conditions, but is over-expressed in cutaneous squamous cell carcinoma (SCC). We previously showed that over-expression of Ets1 in differentiated keratinocytes of the skin leads to significant pro-tumorigenic alterations. Here, we further extend these studies by testing the effects of over-ex...
متن کاملDiagnostic Value of the lncRNA NEAT1 in Peripheral Blood Mononuclear Cells of Patients with Sepsis
Background This study aims to evaluate the diagnostic value of nuclear-enriched abundant transcript 1 (NEAT1) expression in peripheral blood mononuclear cells (PBMCs) for the early diagnosis of sepsis. Methods A total of 59 patients with sepsis, 52 noninfectious SIRS patients, and 56 healthy controls were recruited fort this study. The levels of NEAT1 expression in PBMCs were measured using q...
متن کاملThe oestrogen receptor alpha-regulated lncRNA NEAT1 is a critical modulator of prostate cancer
The androgen receptor (AR) plays a central role in establishing an oncogenic cascade that drives prostate cancer progression. Some prostate cancers escape androgen dependence and are often associated with an aggressive phenotype. The oestrogen receptor alpha (ERα) is expressed in prostate cancers, independent of AR status. However, the role of ERα remains elusive. Using a combination of chromat...
