182 ΔNp63-mediated repression of the lncRNA NEAT1 modulates epidermal differentiation

The transcription factor ΔNp63, a master regulator of epithelial biology, is involved in regulating stem cell function and maintaining the integrity stratified tissues such as epidermis. To this end, ΔNp63 acts transcriptional repressor or activator towards distinct subset protein-coding genes microRNAs. However, how whether regulates long non-coding RNA (lncRNAs) expression not known. Here, we identified lncRNAs NEAT1 bona fide target. We found that proliferating human keratinocytes represses by recruiting histone deacetylates HDAC1/HDAC2 to p63 DNA binding site located proximal promoter genomic locus. Upon induction differentiation, down-regulation associated marked increase levels, resulting an increased assembly paraspeckles foci both vitro skin tissues. RNA-seq analysis with global profile (ChIRP-seq) revealed associates key factors sustaining their during epidermal differentiation. These molecular events might explain inability NEAT1-depleted undergo correct differentiation program.
